- Dipeptidaza E katalizuje hidrolizu dipeptida Asp-Xaa. Ona ne deluje na peptide sa N-terminalnim Glu, Asn ili Gln, niti razlaže izoaspartilne peptide
Slobodna karboksi grupa nije apsolutno neophodna u supstratu.
